FDA-approved adapalene gel is a retinoid labeled for the topical treatment of acne vulgaris in patients 12 years of age and older.1
FDA-approved topical clindamycin products are labeled for the treatment of acne vulgaris.2
Topical nicotinamide (niacinamide) has been studied for inflammatory acne in a randomized comparison with clindamycin gel.3 Tea tree oil from Melaleuca alternifolia has antibacterial, anti-inflammatory, and antioxidant properties that have been reviewed as relevant to acne care.5 In a randomized, placebo-controlled trial, 5% tea tree oil gel was studied for mild to moderate acne vulgaris.4 Across human studies summarized in that review, tea tree oil was associated with fewer inflammatory lesions, mainly papules and pustules, though study designs differed and firm conclusions on overall efficacy remain limited.5 Those studies evaluate the additives on their own and do not establish that this four-ingredient compounded cream is more effective than adapalene or clindamycin alone.
